Posting Summary
Rutgers, The State University of New Jersey is seeking a Postdoctoral Fellow for the Department of Medicine within the New Jersey Medical School.  Under the direction of the Principal Investigator, the Postdoctoral Fellow will investigate the molecular basis of how two Mycobacterium tuberculosis proteins, Rv2623 (a universal stress protein (USP)) and Rv1747 (a putative exporter of lipooligosaccharides) interact to regulate the export of phosphatidyl-myo-inositol mannosides (PIM), immunoregulatory cell wall glycolipids of the tubercle bacillus, thereby influencing anti-TB immunity and modulating in vivo growth of M. tuberculosis. A combination of genetic, biochemical, and immunological approaches, together with various murine experimental tuberculosis models, will be employed to elucidate how the Rv2623-Rv1747-PIM pathway regulate M. tuberculosis growth in the host. The results yielded from the study may illuminate the mechanisms involved in how the growth of M. tuberculosis is regulated in vivo, including in the context of tuberculous latency and reactivation and may lead to the development of anti-TB therapies.
Among the key duties of this position are the following:
  • Generate point mutations of specific genes in M tuberculosis by allelic exchange using the mycobacteriophage-based specialized transduction and site-directed mutagenesis technology.
  •  Conditional knockdown of genes of interest in M. tuberculosis.
  • M. tuberculosis protein-protein interaction assays.
  • Expression and biochemical characterization of M. tuberculosis proteins.
  • Characterization of the in vitro growth kinetics of various M. tuberculosis mutants.
  • Characterization of the in vivo growth kinetics of various M. tuberculosis mutants in a low dose aerosol infection tuberculosis mouse model.

Minimum Education and Experience
  • Doctoral Degree in relevant science.
Required Knowledge, Skills, and Abilities
Preferred Qualifications
  • Experience includes independent design and conduction of genetic and microbiological studies, experience performing unsupervised work in the BSLIII on M. tuberculosis, ability to analyze next generation DNA sequencing data. 
  • Experience in automated, high-throughput fluidic studies of bacterial growth.

Physical Demands and Work Environment
  • This position involves prolonged standing, walking and sitting, as well as use of laboratory equipment. Must perform complex procedures and demonstrate manual dexterity.
  • Occasional lifting of 20 pounds to a height of 6½ feet.
  • Hearing and speech capabilities necessary to communicate with other researchers and staff in person, on the telephone and by e-mail.
  • Vision capable of viewing gauges, computer monitors, charts, forms, text and numbers for prolonged periods.
