Summary

This project is to look at all genes and molecular mechanisms in regulating ATXN1. Among the genes that are validated in vivo in SCA1 mouse brain, the focus will be on transglutaminases that crosslink proteins by a polyglutamine dependent manner. We recently found that transglutaminases preferentially regulate the mutant ATXN1, and reducing the enzyme levels decreases oligomerization of the ATXN1. These results suggests that inhibition of the these enzymes could have therapeutic potential. Based on these findings, the postdoc will study the  molecular mechanisms by which the transglutaminases regulate mutant ATXN1 using in vitro cross-linking/oligomerization assays and using the cell lines that have modified expression levels of the enzymes. This position will also investigate how this enzyme is related to SCA1 pathology in brain histology. Completing this work will enhance our understanding of molecular pathogenesis of SCA1, and will contribute to finding therapeutics for neurodegenerative disorders.

Minimum Qualifications
