Position Highlights The Pond lab within the Department of Cellular & Molecular Medicine is seeking a full-time Research Technician II. The researcher will be responsible for performing basic molecular biology and biochemistry techniques in a laboratory focused on organoid research, including western blotting, cell culture, single-cell analysis, live-cell microscopy, generation of engineered organoid lines, CRISPR screening, spatial RNA sequencing, and advanced image analysis. They may be required to assist with laboratory management including ordering supplies, inventory of biological samples, and maintenance/training of incoming researchers. Researchers will be part of a highly collaborative team of motivated scientists and will be provided with supportive onboarding by experienced professionals. The researcher will be expected to collaborate and mutually support the other lab members to foster a fun and productive lab environment. The researcher is expected to spend 60% of their time doing wet lab experiments and 40% doing image analysis (analysis can be done at a PI-provided workstation or remotely).

Duties & Responsibilities

Duties and Responsibilities:

  • Design, organize, manage, and carry out analytical techniques and subsequent analysis to drive the PI's research. Design research methods in collaboration with senior researchers in Cellular and Molecular Medicine (CMM). Work with researchers to identify and recommend solutions to problems.  
  • Use analyzed data to prepare research reports. 
  • Perform protein expression using gel electrophoresis, western blot, enzyme linked immune sorbent assay (ELISA) recombinant protein expression.
  • Carry out specialized techniques including super-resolution microscopy, electron microscopy and immune hybridization. Study organoid properties from the whole mount level to the single cell level utilizing techniques such as whole muscle testing and permeabilized fiber testing.
  • Assist and train PhD students and postdoctoral fellows to perform and design experiments and on the various techniques utilized in the lab. Direct and assist staff research technicians.
  • Perform bench research including western blotting, RTPCR, extensive cell culture (including generation of primary cultures from patient samples), cell biological assays, and microscopy.
  • Prepare buffers and reagents.
  • Design and carry out single-cell imaging assays.
  • Improve upon current image analysis techniques.
  • Generate live-cell image analysis and collaborate with the UA Nikon center for excellence image analysis group.

Knowledge, Skills & Abilities: 

  • Willingness to learn new imaging and image analysis modalities.
  • Ability to keep protocols, data, and samples organized with clear documentation recording how experiments were performed to ensure rigor and reproducibility.
  • Ability to effectively communicate with professionalism with immediate peers within assigned work group and facilitate interaction and information exchange with other work groups as required to perform role. Based on nature of the role, employee may field requests from others in the organization.
  • High organization skills.
Minimum Qualifications
  • Three (3) years of relevant experience, or equivalent combination of education and work experience required.
  • High school diploma or equivalent, or equivalent learning attained through experience required.
Preferred Qualifications*Microscopy experience. *Tissue culture experience. *Experience curation of data. *Two additional years of relevant laboratory experience.
