Job Description

Job Description

Position Summary

The Janowski Lab discovers and characterizes novel viruses, with a current focus on astrovirus VA1 (VA1). Astroviruses have been long assumed to be pathogens restricted to the gastrointestinal tract, but multiple human patients have been identified to have invasive astrovirus infection of the central nervous system.

We have successfully cultured VA1, the most commonly identified astrovirus species from cases of human encephalitis and developed the first cell culture model that supports replication of VA1 in central nervous system cells. In parallel, we have also developed a murine in vivo model of infection, and surprisingly, VA1 causes myocarditis in mice, suggesting this virus could have additional tissue tropisms beyond the gastrointestinal tract and central nervous system. We are now characterizing the pathogenesis of infection through our cell culture and mouse models of infection. These approaches will allow for important insights into the biology of an emerging family of viruses that cause invasive diseases in humans.

In addition, we are interested in characterizing conserved viral elements between viral families. VA1 contains a highly conserved RNA sequence known as the stem-loop II-like motif (s2m) that is also present in SARS coronaviruses including SARS CoV-2. Currently, the function of this motif is completely unknown despite its presence in multiple viral families. We have developed a reverse genetics system for VA1 and are using it to dissect the function of this motif.

We are seeking an energetic, highly motivated postdoctoral research associate to carry out in vitro and in vivo experiments. Techniques include RNA extraction, PCR, cloning and molecular biology, biochemistry, cell/tissue culture, fluorescence and confocal microscopy, and analysis of RNAseq or other data sets.
